Agrobacterium-Mediated Fungal Resistance Gene Transfer Studies Pertaining to Antibiotic Sensitivity on Cultured Tissues of Lettuce (Lactuca Sativa L. cv. Solan kriti)

چکیده
Lettuce (Lactuca sativa L.) is a widely used leafy vegetable belonging to the family Asteraceae (2n = 18). It is nutritionally rich with medicinal property and well known for its high vitamin A content, and minerals like calcium and iron. This is the only crop which is rich in Lactupicrin and act as an anticancerous as well as, suitable candidate for the production and delivery of therapeutic proteins (Resh, 2001; Ryder, 2002; Mohebodini et al., 2011). However, this crop is severely affected by a number of biotic and abiotic stresses which causes enormous crop yield losses during commercial cultivation of lettuce. Agrobacterium tumefaciens mediated genetic transformation is most common and feasible method to transfer gene of interest into different crop plants and is a widely used method for developing resistance against various diseases (Srivastava, 2003). Agrobacterium-mediated transformation of cotyledonary leaf of lettuce (Lactuca sativa L.) by the GCHI gene
Lactuca sativa is widely used as a leafy vegetable belonging to the Asteraceae family. Cotyledonary leaves of in vitro lettuce seedlings were transformed by plasmids containing the GCHI gene. Also the effects of number of days of pre-cultivation and co-cultivation of explants were examined on the transformation frequency. After co-cultivation, the explants were transferred onto the regeneration...
متن کاملTransfer and expression of the rabbit defensin NP-1 gene in lettuce (Lactuca sativa).
Lettuce (Lactuca sativa L.) is an annual plant of the daisy family, Asteraceae, with high food and medicinal value. However, the crop is susceptible to several viruses that are transmitted by aphids and is highly vulnerable to post-harvest diseases, as well as insect and mammal pests and fungal and bacterial diseases. Here, the rabbit defensin gene NP-1 was transferred into lettuce by Agrobacte...
متن کاملPhosphomannose isomerase gene for selection in lettuce (Lactuca sativa L.) transformation.
A positive selection system using phosphomannose isomerase was employed for Agrobacterium tumefaciens mediated transformation of lettuce (Lactuca sativa L. var. 'Achát'). It was shown that the mannose-based selection system works very well with the lettuce genotype used, reaching up to 25% transformation efficiency on the medium with 20 g/L mannose and 20 g/L sucrose. The best transformation ef...
متن کاملTransformation of Lactuca sativa L. with rol C gene results in increased antioxidant potential and enhanced analgesic, anti-inflammatory and antidepressant activities in vivo
Lettuce is an important edible crop which possesses various medicinal properties. In this study Lactuca sativa L. (cv Grand Rapids) was transformed by Agrobacterium-mediated transformation with rol C gene. Transgene integration and expression was confirmed through PCR and semiquantitative RT-PCR. The transformed extracts were evaluated for their in vitro antioxidant and in vivo analgesic, anti-...
متن کاملEffects of Germination-promoting Substances Given in Conjunction with Red Light on the Phytochrome-mediated Germination of Dormant Lettuce Seeds (Lactuca sativa L.).
Ethylene or thiourea can substitute for gibberellic acid but not for red light in breaking the secondary dormancy induced by extended dark storage of fully hydrated lettuce seeds (Lactuca sativa cv. Grand Rapids). After 10 days of storage, ethylene, thiourea, or gibberellic acid applied either separately or in any combination in conjunction with red light induced near maximal germination. When ...
